pASK-IBA7C is a classic cloning vector devised for E. coli expression of recombinant proteins with the Strep-tagII fused to the N-terminus. It carries the chloramphenicol resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, a multiple cloning site with recognition sites for several common restriction enzymes (e.g. EcoRI, SmaI, BamHI, SalI, PstI), the inducible tetracycline promoter/operator for the regulated expression of proteins, and the sequence for a protease cleavage site.The vector can be combined with any E. coli strain since the tet-promoter works independently of the genetic background of E. coli. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. The recombinant protein will be localized in the cytoplasm. The N-terminal Strep-tagII can be removed by the protease factor Xa.

GCaMP is a genetically encoded calcium indicator (GECI) which was created from a fusion of green fluorescent protein (GFP), calmodulin, and M13, a peptide sequence from myosin light chain kinase. GECI binds Ca2+ and induce a change in fluorescence signal, which allows measuring action potentials and other receptor activation events that trigger Ca2+ fluxes. In addition to the direct cloning of the gene of interest into pASG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

A mutated form of estrogen ligand-binding domain (ERT2) that binds to synthetic antagonists (such as tamoxifen or its derivative 4-hydroxy-tamoxifen) but not to circulating estrogens was fused to the Cre recombinase (Cre) to create the CreERT2, which requires the presence of tamoxifen for Cre recombinase activity. Duplex DNA is isolated from bacteriophage lambda (cI857ind 1 Sam 7). The bacteriophage is grown in an E. coli host which is deficient in adenine methylase (dam-). Therefore, those deoxyadenosine positions which are methylated in a wild type E. coli host are not methylated in the dam- host. This DNA gives complete digests with restriction endonucleases that are sensitive to dam methylation. Lambda DNA (dam-) is 48,502 base pairs in length.
